GHRP-6

Growth Hormone Releasing Peptide-6

The original GH releaser — and the hungriest

A first-generation hexapeptide ghrelin-receptor agonist that drives a strong growth hormone pulse and a pronounced surge in appetite. GHRP-6 is the most ghrelin-like of the GH secretagogues, which makes it a favorite for hard-gainers and recovery — but also the one most likely to raise cortisol and prolactin at higher doses.

Mechanism of Action

How GHRP-6 works

GHRP-6 mimics ghrelin to release growth hormone through the GHS-R1a pathway:

  • Ghrelin receptor (GHS-R1a) activation — binds the growth hormone secretagogue receptor on pituitary somatotrophs, triggering a calcium-dependent GH pulse. As a true ghrelin mimetic it activates the receptor strongly but non-selectively.
  • Marked appetite stimulation — the same ghrelin signaling that releases GH acts on the hypothalamic arcuate nucleus to sharply increase hunger, often within 20–30 minutes. This is GHRP-6's signature effect and distinguishes it from the cleaner Ipamorelin.
  • Broader hormonal cascade — at higher doses it can mildly raise cortisol, ACTH and prolactin, unlike the highly selective Ipamorelin. Dose discipline keeps these off-target effects minimal.
  • Cytoprotective signaling — GHS-R1a activation has documented gastric and cardiac cytoprotective effects in research models, independent of the GH axis.

Your Genetics & GHRP-6

Genetic variants that affect your response

These SNPs determine how effectively GHRP-6 works for you specifically. A genetic peptide report identifies your variants before you start.

GHSRrs572169
Ghrelin receptor sensitivity

Variants in the growth hormone secretagogue receptor gene change receptor density and signaling efficiency. Lower GHSR expression means fewer targets for GHRP-6, blunting both the GH pulse and the appetite response.

GHRLrs696217
Endogenous ghrelin (Leu72Met)

The Leu72Met variant alters ghrelin processing and circulating levels and is linked to differences in appetite and GH secretion. Since GHRP-6 acts on the same receptor as your own ghrelin, this variant shapes how strong the hunger surge feels.

GHRrs6873545
Growth hormone receptor (d3/fl polymorphism)

The GHR exon 3 deletion (d3-GHR) increases downstream GH sensitivity. d3 carriers convert each GHRP-6 GH pulse into greater IGF-1 output, often reaching the same result at a lower dose.

SSTrs4988514
Somatostatin (GH inhibition)

Somatostatin is the brake on GH release. Variants that raise somatostatin tone can partially blunt the GHRP-6 pulse, making timing (dosing when somatostatin is naturally lower, such as the evening) more important.

Sourcing & access

Where to buy GHRP-6

GHRP-6 is sold as a research compound, not a licensed medicine, so quality and legal status vary widely by country and vendor. If you're sourcing it, treat independent purity testing as non-negotiable.

  • Insist on a recent third-party Certificate of Analysis (COA) for the exact batch — labs like Janoshik verify purity and identity.
  • Check the legal status where you live before ordering — it differs from country to country.
  • Reconstitute correctly with bacteriostatic water and start at a conservative dose.

Swiss Chems is a US research-chemical supplier that publishes third-party Certificates of Analysis. Sold for research use only — check your local laws.

CJC-1295

GH Synergy StackModerate Evidence

CJC-1295 (Modified GRF 1-29)

A 29-amino-acid growth hormone releasing hormone (GHRH) analog with enhanced stability. CJC-1295 extends the duration of natural GH pulses without disrupting pulsatile release patterns, providing sustained elevation of growth hormone and IGF-1 for recovery, body composition, and anti-aging.

Ipamorelin

cleaner GH pulse alternativeModerate Evidence

Ipamorelin Acetate

A pentapeptide ghrelin receptor agonist that triggers clean, selective growth hormone pulses without raising cortisol or prolactin. Ipamorelin is considered the most selective GH secretagogue available, making it the preferred choice for those prioritizing minimal side effects alongside GH optimization.

Frequently Asked Questions

What is GHRP-6 used for?

GHRP-6 is used to stimulate natural growth hormone release for body composition, recovery and sleep — and notably to increase appetite. Its strong ghrelin-mimetic action makes it popular with hard-gainers, but it is less selective than Ipamorelin and can raise cortisol and prolactin at higher doses.

Why does GHRP-6 make you so hungry?

GHRP-6 acts on the same receptor as ghrelin, your body's hunger hormone. Activating GHS-R1a in the hypothalamus triggers a sharp appetite increase, usually within 20–30 minutes of dosing. The intensity varies by genetics — GHRL and GHSR variants influence how strong the effect is.

Does genetics affect GHRP-6 response?

Yes. GHSR variants set how many ghrelin receptors GHRP-6 can activate, the GHRL Leu72Met variant shapes the appetite and GH response, the GHR d3 polymorphism affects downstream GH sensitivity, and somatostatin variants influence how strongly your body counteracts the pulse. A genetic peptide report profiles these for dose optimization.
